Distinct molecular properties and functions of small EV subpopulations isolated from human umbilical cord MSCs using tangential flow filtration combined with size exclusion chromatography

This study establishes a protocol for large-scale production of MSC-derived sEV subpopulations from human umbilical cord MSCs using TFF combined with SEC. The resulting S1-sEVs (CD9/HRS/GPC1 enriched) showed greater immunomodulatory impact, while S2-sEVs (CD63/FLOT1/2 enriched) showed enhanced capacities in promoting cell proliferation and angiogenesis—demonstrating distinct functions both in vitro and in animal disease models.
